Summary: We performed single-cell RNA-sequencing (scRNAseq) on 14 human medulloblastoma patient samples spanning all subtypes. We used this dataset to uncover novel interactions between the tumour, and its microenvironment, that promotes medulloblastoma progression and metastasis. We have identified the presence of 10 distinct populations in the global view including GNPs, Oligodendrocyte Precursor Cells (OPCs), Oligodendrocyte, Myeloid cells, and Lymphocytes. One of the populations analyzed in detail were the myeloid cells, where both, the resident microglia, and the bone-marrow-derived macrophages were identified and characterized. In addition, a transitionary cluster formed between tumour cells and myeloid cells, that expressed high levels of both neuronal and immune markers was identified and was predicted to include tumour cells and granule neuron precursors (GNPs) being phagocytosed by myeloid cells.
