Abstract
"The cell membrane is an effective barrier hindering the transport of molecules into or out of the cell. Nanostraws are hollow nanotubes capable of piercing the cell membrane, resulting in a direct fluidic pathway into the cytosol. The membrane piercing is of a general nature and works both for model cell lines and high value primary cells. The method is exceptionally gentle and do not affect cell viability, proliferation, or gene expression.
In this talk, I will describe the nanostraws and showcase how they can be used to both introduce exogenous molecules to cells (transfection) and to extract cellular contents (sampling). For the transfection, I will show high efficiency introduction of nucleic acids to primary human hematopoietic stem cells. For the sampling, I will show repeated, longitudinal extraction of mRNA tracking cellular transformations."
